We created hybrids between Saccharomyces cerevisiae (Sc) and Kluyveromyces marxianus (Km) by removing telomeres from Sc chromosome I or III for circularization and adding Km centromere and replication origins. The engineered chromosomes I and III were individually extracted from Sc and transferred into Km, each resulting in a hybrid containing eight natural Km chromosomes and one Sc chromosome. Overnight cultures of three parallel samples of parental and hybrid strains were diluted into YPD to achieve an OD600 of 0.2. Once the OD600 of the cultures reached 0.6, cells were collected directly for the YPD group. For stress treatment, the culture was supplemented with tunicamycin (TM) at a final concentration of 1 mug mL-1, or with NaCl at a final concentration of 1 M, and cells were collected after 1 hour.
