Gene expression profile at single cell level of islet cells from normoglycemic or hyperglycemic mice

The purpose of the experiment was to investigate the heterogeneity of islet cell response, especially beta-cells to hyperglycemia induced by severe insulin resistance in mice. Mice were implanted with osmotic pumps containing PBS or S961 insulin receptor antagonist (12nM). S961-treated mice developed hyperglycemia within 24h. Pancreatic islets were isolated 5 days after transplantation, dissociated into single cell suspension and stained for propidium iodide. Propidium iodide-negative live cells were FACS-sorted and single cells captured in droplet emulsions to generate libraries using a Chromium Single-cell instrument (10X Genomics).
