Chronic inflammation-related diseases are chronic conditions marked by persistent tissue damage and slow healing, which can progress to irreversible deterioration and high mortality. Osteoarthritis (OA) is a common manifestation of chronic inflammatory diseases. Current small molecule drugs are inadequate for treating osteoarthritis. Polyphenols, long-recognized for their anti-inflammatory and antioxidant properties, have limited effectiveness due to their short half-life and low bioavailability. We have developed a simple strategy to create metal-free polyphenol capsules that prolong bioactivity and enhance bioavailability. These capsules effectively scavenge reactive oxygen and nitrogen species and reduce intracellular ROS and inflammatory markers in various cell models. They also inhibit the p38 MAPK signaling pathway, which is crucial in inflammation associated with osteoarthritis. Our research provides a simple and efficient strategy for polyphenol-based therapeutics with significant potential for the treatment of osteoarthritis in the clinic.
