In this study, we analyze the differential binding pattern of Nac1, a pluripotency transcription factor, in mouse embryonic stem (ES) cells and in their differentiated states - mesendoderm (ME) and neuroectoderm (NE) cells. ES cells were maintained in DMEM medium containing LIF and BMP4. While ES cells were differentiated into ME using Chiron (Wnt agonist)and activin-A, they were differentiated into NE using retinoic acid. We performed chromatin-immunoprecipitation ChIP)of Nac1 in ES, ME and NE cells followed by next-generation sequencing of the Nac1 bound DNA.
