A Sleeping Beauty transposon-based forward genetic screen was performed to identify the genes that promote osteosarcoma development. Mutagenesis was performed in wild type and Trp53 deficient animals. Animals were aged until tumors developed or until moribund. Upon necropsy, tumors were divided into 3 portions and snap frozen for DNA extraction, fixed in 10% formalin for histological analysis, and snap frozen in RNAlater (Sigma) solution for RNA extraction. Pathological analysis and tumor classification was performed at the National Institute of Health / National Cancer Institute. Ten of these tumors were selected for RNA-Sequencing.
