Overexpression of transgenic rat prolactin ligand in mammary epithelial cells results in dysplasia followed by spontaneous development of ER positive mammary tumors, referred to as the NRL-PRL model. This study generated whole genome and exome sequencing of five mammary tumors and matched normal tail tissue from aged female NRL-PRL mice in an effort to identify the mutational events leading to tumor progression. RNA sequencing was performed on these five tumors and adjacent dysplastic mammary tissue, in addition to five cell preparations developed from nondysplastic mammary epithelial cells from young NRL-PRL females, in order to further elucidate the signaling events associated with tumor establishment. Activating mutations in Kras were identified across this Discovery Set of tumors. This study also generated whole exome sequencing of eight additional tumors (three had matched tails) to discover additional somatic alterations in Kras-wildtype NRL-PRL tumors.
