Time-lapse confocal (LSM 780, Carl Zeiss) images of COS-7 cells were acquired every 30 s over 22 min through a 63x 1.45NA oil-immersion objective. Rapamycin was added after frame 5. Four fields were acquired in parallel from 4 different locations on the same coverslip using the motorized stage. This is field #2, which is shown in the published figure. Cyan, CFP-FKBP-PIP5K; magenta, autofluorescence (iRFP channel, with no iRFP-tagged protein expressed in these cells); green, GFP-P4M; red, FRB-mCherry-giantin(tail); white, DIC.
