Promoter-Enhancer Communication Occurs Primarily within Insulated Neighborhoods

Esrrb was shown previously to interact directly with Mediator in mouse embryonic stem cells (mESCs) and Esrrb depletion does not lead to mESC differentiation. Based on these facts, we used Esrrb knockdown (Esrrb KD) as a proxy for enhancer inactivation, because Esrrb KD removes Mediator from the enhancers. These Western blots show that Esrrb indeed recruits Mediator to DNA template and Mediator plays a key role in assembling pre-initiation complex (PIC).
