Provided are the isolation and cloning of gene orf188 capable of regulating the oil content and cytoplasmic effect of rapeseed and an application thereof. The gene orf188 is located in the mitochondrial genome of the rapeseed and is unique to some mitochondrial genomes of the rapeseed. The gene orf188 has the function of regulating the oil content of the rapeseed. Increasing the expression of the gene orf188 by means of genetic engineering technology may increase the oil content of the rapeseed， and the purpose of increasing oil production is thus achieved.
