We performed RNA-seq of livers obtained from mice fed either a control or the NASH-inducing FPC diet (Wang et al., 2016) for 24wk. In some FPC-fed mice the NASH-activated transcription factors, Elf3 or Glis2, had been deleted specifically in hepatocytes, when mice reached a steatotic state. HEP-INTACT mice were fed either the FPC diet or a control diet for 14wks. Mice were tail-vein injected with either AAV2/8-LP1-GFPmut-miRNA_NC, AAV2/8-LP1-GFPmut-miRNA_Elf3 or AAV2/8-LP1-GFPmut-miRNA_Glis2 (5*1011 p.f.u./mouse) and mice were kept additionally 10wks on the diet before livers were harvested for RNA-seq analyses.
