We placed immotile variants of the soil bacterium P. fluorescens SBW25 (SBW25(delta)fleQ IS-(omega)Km-hah: PFLU2552) under selection for recovered flagella-mediated motility using soft agar as a directional selection medium. We observed that typically a single nucleotide polymorphism (SNP) would arise de novo within 72-120 hours and restore the motility phenotype. The most frequently observed mutation (observed >95% of independent replicates) was a single SNP in the ntrB gene encoding the histidine kinase NtrB (mutation ntrB A289C). A subset of these evolved lines was sent for whole-genome sequencing to screen for secondary, background and hitchhiking mutations.
