2nd passage (P2) recombinant baculovirus vectors having a reporter green fluorescent protein (mAzami Green) under the control of the p6.9 promoter. The recombinant viruses were amplified in Spodoptera frugiperda (Sf9) cells at a low multiplicity of infection (MOI), ~0.1, and harvested 72 hours post infection (hpi). The baculovirus genomic DNAs (gDNAs) from the propagated P2 p6.9GFP rBEV stock or P2 p6.9GFP_sgRNA_gp64+131 rBEV stock were extracted and sequenced via shotgun sequencing or tiled-amplicon sequencing, respectively. Control and CRISPR-Cas9 targeted baculovirus genomes, upon plasmid or rBEV-based delivery of sgRNA into Sf9-Cas9 cells, were extracted and sequenced via tiled-amplicon sequencing.
