Gonadotrophin releasing hormone (GnRH) from hypothalamus stimulates gonadotrophin secretion from pituitary, followed by the secretion of sex steroid hormones from ovaries. GnRH is also detected in the extra-hypothalamic areas, like trophoblasts at the implantation sites. We have previously studied the association between trophoblasts and mast cells by producing leukemia inhibitory factor (LIF) and matrix metalloprotease (MMP)-9, and here hypothesized another association between the cells via GnRH derived from trophoblasts. We here report the RNA-seq analysis revealed the expressions of LIF, MMP-9, and a NK cell attractor CXCL16 enhanced in a human mast cell line LAD2 with forced-expression of GnRHR (GnRHR-expressing LAD2), when compared to those of control LAD2. Lentivirus was used to force expression of GnRHR or EGFP as a control in LAD2 cells. The GnRH agonist, leuprorelin, or DMSO was added, and cells were cultured for 72 h. Total RNA was then extracted for RNA-seq profiling.
