A microfluidic strategy is described to simultaneously capture, purify, and sequence TCR genes from CD8+ T cells, while labeling those genes with DNA barcodes that encode the peptide sequence of the cognate antigen as presented by MHC complexes. This establishes the link between peptide antigens and TCR sequencing in a simple microfluidic device, to analyze rare blood cells in limited patient samples without requiring complex FACS sorters and equipment.
