To study the transcriptome in developing heart controlled by the intact Mediator complex. RNA-seq analysis was performed on RNA extracted from hearts of Med30 cKO and control littermates at E10.5. From this analysis, 10,637 genes were expressed in both mutant and control cardiomyocytes. Of these, 31 genes were significantly upregulated, and 45 genes were significantly downregulated (log2fold > 1, adjusted p value < 0.05) in mutant cardiomyocyte relative to controls. To identify genes responsible for the rapidly developed phenotype in Med30 icKO mice, we performed RNA-seq analysis of RNA extracted from adult cardiomyocytes isolated from Med30 icKO and control mice at 4-weeks post tamoxifen injection. From this analysis, 9517 genes were detected in both mutant and control cardiomyocytes.
