Bacillus thuringiensis as a Gram-positive soil bacterium, is a member of the B. cereus group. B. thuringiensis (Bt) possesses crystal-like parasporal proteinaceous inclusions which have unique and selective biocidal activities on different insects, nematodes, and human cancer cells. The project study was conducted to identify the parasporal inclusion proteins that had in vitro cytotoxicity to the HeLa cancer cell line. B. thuringiensis strain 28s was isolated and identified from a soil sample of the Narsingdi district of Bangladesh. B. thuringiensis strain 28s produces about 0.8 micrometer sizes of round-shaped parasporal inclusion protein. When this inclusion protein was solubilized with 53 mM freshly prepared sodium carbonate solution at pH 11.0, it produced 3 different sizes (91 kDa, 86 kDa, and 76 kDa) protein bands in SDS-PAGE. When the solubilized parasporal inclusion proteins of B. thuringiensis strains 28s were incubated for the proteolytic digestion with proteinase K, and trypsin in separate microcentrifuge tubes at 37oC for 1 hour in a water bath, a ~46 kDa proteinase K digested single protein fragment was produced and 4 different sizes, ~51 kDa, ~45 kDa, ~42 kDa, and ~32 kDa protein fragments were also produced from trypsin digestion. Both proteinase K and trypsin-digested protein fragments showed strong cytotoxicity to the human cervical cancer cell line (HeLa). The project of whole genome sequencing of B. thuringiensis strain 28s was therefore undertaken to reveal the potential insights of the bacterium.
