Bulk RNA-seq of microglia from Apoa1 KO mice injected with ApoA-I protein intravenously

We found ApoA-I from blood can be taken up by microglia in the brain. To determine if ApoA-I from circulation is sufficient to restore microglial transcriptomic state to a more wild-type-like state, we performed a rescue experiment by intravenously injecting ApoA-I protein into adult Apoa1 KO mice and sorted microglia with or without ApoA-I uptake for bulk transcriptomic analysis. Overall design: We intravenously injected ApoA-I protein three times, two days apart, into adult Apoa1 KO mice. One day before taking down the mice, we injected dye-labeled ApoA-I into both wild-type mice and Apoa1 KO mice. We then sorted ApoA1-Pos and ApoA1-Neg microglia from both WT mice and Apoa1 KO mice for bulk RNA-seq analysis. We have 4 groups in total and 4 replicates per groups.
