The study explores the interplay between the T-cell response, microbiota, and Parkinson's Disease (PD). PD involves neuroinflammation, motor impairment, and pain, primarily driven by alpha-synuclein aggregation. Research suggests alpha-synuclein as a source of neoantigens, eliciting autoreactive T-cell responses. However, the link between microbiota and this response remains unclear. Using transgenic SNCA mice, the study depleted microbiota and lymphocytes to assess their impact on PD symptoms. Results show microbiota and T-cell depletion alleviating motor deficits, sensory impairment, and gut inflammation. Additionally, a T-cell response to alpha-synuclein-derived antigens is identified, possibly induced by molecular mimicry with gut bacteria. This highlights the role of T-cells and microbiota in PD progression.
