The selective regulations underlying these two roles in the liver remain to be quantitatively explored. We measured liver gene expression after oral glucose administration of 5 different glucose doses (0.25, 0.5, 1, 2, 4 g/kg-weight) and water administration in WT mice. We fasted 10-week-old WT mice for 16h and orally administrated 5 different glucose doses or water. We collected livers and blood samples at 0, 20, 60, 120, and 240 min after the administration. RNAeasy Mini Kit (QIAGEN) and Qiashredder (QIAGEN) were used to extract total RNA from 10 mg of liver samples following to the protocol. The quantity and the quality of the extracted RNA were assessed by Nanodrop (Thermo Fisher Scientific) and the 2100 Bioanalyzer (Agilent Technologies). The sequencing library was prepared as previously described and subjected to sequencing using HiSeq 2500 (Illumina).
