This data is a part of our study that suggests Wnt/GSK-3β mediates posttranslational modifications of FLYWCH1, regulating intestinal epithelial function and tumorigenesis in the colon. TMA Immunohistochemistry (IHC) analysis Immunohistochemistry for FLYWCH1 (1:500; Atlas Antibodies, HPA040753) and GSK-3B (1:400; Cell Signaling, 12456) was conducted in collaboration with the Histopathology Unit at City Hospital, University of Nottingham, using a CRC TMA containing 1000 primary colorectal cancers [6]. The TMA was generated from resection specimens’ of 1000 patients with CRC at City Hospital, University of Nottingham, who underwent surgery between 2008 and 2012. The average age of the patients was 68 years, and the mean follow-up time was 53 months. After completing the immunohistochemistry, the TMA slides were scanned at 20x magnification using the VENTANA DP 200 slide scanner (Roche, UK). The staining intensity was scored using the H-scoring system, calculated as previously described [7]. TMA-stained sections were first checked by W.D. and E.R. to ensure the areas were defined correctly before scoring. Furthermore, cores with less than 15% invasive tumour tissue or intact (epithelial) tissue were excluded (i.e., not scored). The H-score ranged from 0 to 300, using the formula: (1 × % weakly stained nuclei) + (2 × % moderately stained nuclei) + (3 × % strongly stained nuclei).
