Gene expression profile on pons and medulla of maternal immune activation model rat

We created two-hit immune activation model by inducing a pseudo-bacterial infection (administration of lipopolysaccharide (LPS)) on postnatal (P) 11 in pups born to rats infected with a pseudo-virus (administration of polyinosinic-polycytidilic acid (poly I:C)) during pregnancy of day 10. Our model rats showed increased mortality rate due to LPS administration on P11 and abnormal blood biochemical test results. Next, we examined the gene expression in pons and medulla oblongata of our model rats on P11 by DNA microarray and found that a number of genes showed more than a 2-fold change in expression.
本研究构建了二次打击免疫激活模型:对妊娠第10天通过注射聚肌胞苷酸(poly I:C)感染模拟病毒的母鼠所产幼崽,于出生后第11天(P11)诱导模拟细菌感染(通过注射脂多糖(LPS))。该模型大鼠在P11接受LPS注射后死亡率升高,且血液生化检测结果出现异常。随后,本研究通过基因芯片(DNA微阵列)检测了模型大鼠P11时刻脑桥与延髓的基因表达情况,发现大量基因的表达量变化幅度超过2倍。
