Investigation of the Impact of Microbial Communities and Soil Physicochemical Properties on the Successful Cultivation of Morchella in Greenhouses

In April 2024, some greenhouses in the morel mushroom cultivation park in Shuangcheng District, Harbin City, Heilongjiang Province, China, experienced dead mushrooms and no mushrooms. To investigate the reasons for these issues, we used a five point sampling method to sample the surface soil (1-5cm) in the same greenhouse and mixed these soil samples into one sample. The soil samples from each location were sieved through a 2mm sieve and roots and plant debris were removed by hand. Subsequently, we divided the sample into two parts, one part was air dried for soil physicochemical property determination, and the other part was quickly placed in liquid nitrogen, brought back to the laboratory, and stored in a -80 Celsius freezer for subsequent DNA extraction. A total of 9 soil samples were collected this time, including normal mushroom samples (CK, 3 samples), dead mushroom samples (DM, 3 samples), and non mushroom samples (UM, 3 samples)
