We performed single-cell RNAseq and bulk RNAseq analysis to investigate the function of CD133 and a new mechanism of intercellular comminication for cell proliferation, in mouse liver, following partial hepatectomy (PHx). Overall design: For single-cell RNAseq, hepatocytes were isolated from wild type (WT) and hepatocyte-specific Shp2 knockout (SKO) livers at 4 hours and 2 days following PHx, and Shp2/Prom1 double knockout (DKO) livers at 2 days following PHx. For bulk RNA-seq, total RNA was isolated from CD133+ vesicle, extracellular vesicle and whole cells.
