Breast cancer is the most common malignancy among women worldwide. Medical breakthroughs have increased treatment options and survival for breast cancers; yet, the incidence of breast cancers remains on the rise. Life style-induced metabolic syndrome, such as diabetes and obesity, posed to be a major health concern and have been associated with increased cancer risks. We hypothesize that the accumulation of toxic metabolites in breast tissues may provoke carcinogenesis by enhancing the rate of mutagenesis. If uncorrected, accumulated damage may lead to excessive DNA damage and trigger malignant transformation. Work done by us and others have demonstrated that intracellular metabolites may lead to increased mutagenesis. In this study, we developed in vitro culture treated with methylglyoxal to look for mutational signatures in mammary cells and organoids.
