This study performed single-cell RNA sequencing (scRNA-seq) analyses on gene transcriptions of parental Wild-type and F508del-CF HL60 cell lines, and their DMSO-derived neutrophils with or without LPS stimulation. There are 6 groups of cells subjected to scRNA-seq: 1) Parental wild-type HL-60 cell line (WT_HL-60); 2) Parental F508del-CF HL-60 cell line (CF_HL-60); 3) Neutrophils differentiated from wild-type HL-60 cell line (WT_dHL-60); 4) Neutrophils differentiated from CF-F508del HL-60 cell line (CF_dHL-60); 5) WT_dHL-60 stimulated with LPS (WT_dHL-60_LPS); and 6) CF_dHL-60 stimulated with LPS (CF_dHL-60_LPS). For differentiation induction, 1.25% DMSO was added to the complete RPMI-1640 culture medium for 5 days with a medium replenishment at Day 3. For LPS stimulation, Pseudomonas aeruginosa LPS (42.67 ug/ml) was added for 16 hours.
