Despite being intestine-restricted, the helminth Heligmosomoidese polygyrus bakeri (Hpb) induces systemic accumulation of Th2 cells, providing the opportunity to sequence Th2 cells in multiple tissue of the same mouse. Here we first compared IL-5-Cre and IL-13-Cre for permanent fluorescent labeling of Th2 effector cells after Hpb infection Next, we compared IL-5-Cre traced memory Th2 cells from multiple tissues: mesenteric lymph nodes (MLN), small intestine lamina propria (SILP) and mesenteric fat (MFAT). As a control, we also sorted naive CD4+ T cells from the MLN. Overall design: We bred Red5 (RFP and Cre expressed from the Il5 locus) and YetCRE13 (YFP and Cre expressed from the Il13 locus) mice with Ai6 (Rosa26::STOPfl/fl::ZsGreen) or Ai9 (Rosa26::STOPfl/fl::TdTomato) mice, respectively, to generate lineage traced Il5-Ai6 and Il13-Ai9 mice.These mice were infected with Hpb and drug cleared with pyrantel pamoate after 14 days. Cells were sorted by flow cytometry 4 weeks after drug clearance.
