Himar1 TnSeq profiling of M. tuberculosis H37Rv grown on oleate and/or glycerol at acidic and neutral pH
收藏资源简介:
Mtb H37Rv Himar1 transposon-mutant (TnSeq) library grown in 7H9 with oleic acid vs glycerol at pH 5 vs pH 7 to identify genes required for growth in acidic conditions, which depends on presence of fatty acids in medium. Data collected by Alex Gouzy and Claire Frances Healy (Sabine Ehrt lab, Weill Cornell Medical College, NYC).Medium used:Base medium used: 7H9-AN (FAF-BSA) + tyloxapol 0.05% + Kana 50ug/mLAN= Albumin + NaClFAF-BSA= Fatty acid free bovine serum albuminCondition tested: Conditions tested varied depending on the carbon source provided (glycerol or oleic acid) and pH (pH7 or pH5)5 conditions in total:pH7 Glycerol 0.2%pH7 Glycerol 0.2% + OA 200uMpH7 OA 200uMpH5 Glycerol 0.2% + OA 200uMpH5 OA 200uMFor comparison, 'Input' samples are included (grown in 7H9-ADN (albumin+dextrose+NaCL), pH ~7, carbon source: glucose+glycerol).2 passages (P1 and P2, effects are stronger in P2).3 replicates of each condition (exp1-3).
