Global gene expression profiling of SM and the derivatives (DM, D, MYO, SCL, SYN) induced from hiPSCs

Global gene expression profiling of human iPSC and the iPSC-derived presomitic mesoderm(PSM), somite(SM), and the derivatives, dermomyotome(DM), dermatome(D), myotome(MYO), sclerotome(SCL) and syndetome(SYN). Total RNA was prepared using the RNeasy Mini Kit (Qiagen). cDNA was synthesized using the GeneChip WT (Whole Transcript) Sense Target Labeling and Control Reagents Kit as described by the manufacturer (Affymetrix, Santa Clara, CA). Hybridization to the GeneChip Human Gene 1.0 ST expression arrays, washing, and scanning were performed according to the manufacturer’s protocol (Affymetrix). Expression values were calculated using the RMA summarization method, and the data obtained were analyzed by GeneSpring GX 14.5 (Agilent Technologies, Santa Clara, CA, USA).
