Within this study, we address the impact of spatial variability on the diversity of peatland methanogens and methanotrophs, in order to first determine the functional genetic diversity by applying captured metagenomics on genes encoding for enzymes related to metabolism of methane producing and consuming microorganisms. We apply the captured metagenomics method to nine peat mesocosms with three different sampling locations to test the following hypothesis and aims. (1) Does the frequency of fwdX, ACDS and pmoA genes have a positive effect on the magnitude of CH4 flux; we aim to (2) determine whether the functional diversity increases with depth in a peat profile; and finally (3) identify whether the 13C of CH4 match the dominant genes in the methanogenesis pathway.
