Abstract Dengue infection is the fastest spreading mosquito borne disease in the world. More than 2.5billion people spread over more than 100 countries are at risk. Thrombocytopenia is a key clinical feature and around 500,000 people annually develop the more severe form of this infection known as severe dengue (Dengue Haemorrhagic Fever/ Dengue Shock Syndrome). Platelets are key in the pathogenesis of severe dengue. Platelets have a cardinal role in haemostatic function and innate immunity, both of which are compromised in severe dengue. There are currently no drug treatments or vaccines available for dengue infection. The review highlights the role of platelets in humans and integrates the pathology seen in dengue with the known physiology. It also gives an insight on the drug targets being researched and why platelets should be considered as a drug target in dengue haemorrhagic fever.
