These .fcs files report the effect of different carbon sources/growth environments (Glucose, Galactose, Glycerol, Ethanol) on the expression level of TDH3 promoter variants in the model yeast Saccharomyces cerevisiae. TDH3 promoter alleles were fused to the yellow fluorescent protein (YFP) and the reporter constructs were genomically integrated into the HO locus to create transgenic strains. Expression levels of the transgenic strains were quantified using flow cytometry.
Conclusion:
The promoter variants used in this study spanned an expression range from ~0% to ~200%. The vast majority of promoter variants had different expression levels in different environments. Samples were grown and scored in four replicate 96 well plates, for a total of 384 samples. Each plate contained 24 replicates of a control strain containing a wild type allele of the TDH3 promoter driving expression of YFP. These controls can be used to correct for technical variation between plates or flow cytrometry runs.
