Collectively, our study reveals the role of Olig2 in hindering the in vivo AtN conversion and idendifies an inducible barrier to neuronal reprogramming. Overall design: WT mouse cortex was injected with AAV9 hGFAP-Ngn2-tdTomato. Virus-infected brain tissue was collected 7 days post injection (n = 12 mice), and dissociated into single-cell suspension, followed by tdTomato+ cells collecting by fluorescent-assisted cell sorting. The tdTomato+ cells were then subjected to CUT&Tag sequencing.
