A method to enrich centromeric DNA from human cells

We have developed a technique based on selective restriction digestion and size fractionation to enrich for centromeric DNA from human cells. Combining enzymes capable of cutting at high frequency throughout the genome, except within most human centromeres, a highly enriched sample was obtained. This will provide applications for targeted sequencing of centromeres with short or long-read technology as well as direct visualization of single molecules.
