This study investigated the transcriptomic changes in human THP-1 macrophages in response to Candida albicans SC5314 infection, with or without cinnamaldehyde treatment. RNA-seq revealed 1,996 differentially expressed genes (DEGs) (|log2FC |= 1, adjusted p-value < 0.05), comprising 691 upregulated and 1,305 downregulated genes. Additionally, we provide RNA sequencing data for macrophages uninfected with C. albicans. Between macrophages infected with C. albicans and uninfected macrophages, a total of 18 DEGs were identified, with 15 upregulated and 3 downregulated. Overall design: Plate THP-1 cells at a density of 8 à 105 cells/mL in a 6-well plate. Induce differentiation with 100 ng/mL PMA for 24 h. After cells adhere, add Candida albicans (final concentration: 2.4 à 106 CFU/mL) and cinnamaldehyde (final concentration: 62.5 µg/mL), then incubate for 2 h. Following incubation, aspirate the medium. Wash cells by slowly adding 1 mL of ice-cold PBS down the side of each well, gently rocking the plate for a few seconds, and aspirating the PBS. Add 500 µL of TRIzol reagent to each well, pipette to homogenize, and transfer the lysate to pre-chilled RNase-free cryovials.
