Myeloid-derived suppressor cells (MDSC) are major negative regulators of immune responses in cancer and chronic infections. It remains unclear if regulation of MDSC activity at different conditions is controlled by similar mechanisms. In order to compare MDSC in mice with cancer and lymphocytic choriomeningitis virus (LCMV) infection, we would like to perform gene profiling and comparison of M-MDSCs in tumor bearing and LCMV infected mice using total RNAseq: Overall design: Sort and fix M-MDSC isolated from spleen in 1) naïve mice, 2) LLC tumor bearing mice on 3 weeks, and 3) LCMV infected mice on 2 weeks, 2. extract RNA using a FFPE isolation ki (QIAGEN) with Rnase, 3. Run total RNAseq, 4. Analyze data and compare gene expression between LLC and LCMV mice.
