Genome-wide DNA methylation in cultured human Schelmm's Canal endothelial cells and trabecular meshwork cells with or without glaucoma

Genome-wide DNA methylation profiling in cultured human Schelmm's Canal endothelial cells (SC) and trabecular meshwork (TM) cells derived from post-mortem eyes with or without glaucoma. Our study aimed to identify glaucoma-associated genes that were affected by DNA methylation. DNA were extracted from cultured human SC (n=9, 2 glaucoma, 7 non-glaucoma) and TM (n=5, 1 glaucoma, 4 non-glaucoma) cells using Allprep DNA Mini spin column kit. DNA samples were hybridized to the Illumina Infinium HumanMethylation450 Beadchips. Probe and intensity data were normalized and analyzed by RnBeads package in the R program.FDb.InfiniumMethylaion.hg19 package was used to annotate probes. The selected genes were validated using Zymo qMethyl kit.HumanMethylation450 Beadchips.
