RNA-seq analysis of human iPSC derived hepatocyte and stellate cell surround organoids (iHSOs)

To investigate the interaction between hepatocytes and hepatic stellate cells in iHSOs, we established contact co-culture organoids using iPSC-derived HSCs and hepatocytes; termed iPSC-derived hepatocyte-stellate cell surround organoids (iHSOs). Sorting based on fluorescent labeling (GFP/tdTomato) of the iPSCs used in iHSOs allowed analysis of each cell type. A comprehensive RNA-seq analysis was conducted. In addition to preparing RNA-seq samples from hepatocytes and HSCs sorted from iHSOs using FACS (CoSp-SortHep, n=3; CoSp-SortHSC, n=3), we also prepared RNA-seq samples from iPSCs (n=1), iPS-Heps (n=2), iPS-HSCs (n=2), iPS-Hep mono-spheroids (iPS-Hep-Sph, n=2), and iPS-HSC mono-spheroids (iPS-HSC-Sph, n=2).
