we performed the RNA-seq and de novo assembly of leaf transcriptome of 16 Nicotiana species and identified the phylogenetic relationship of these Nicotiana species on the transcriptome levelusing the Maximum Likelihood method.Furthermore, the expression profiles of the 16 species were calculated and the Person correlation analyses were conducted.This is the first report on the leaf transcriptomes of sixteen Nicotiana species by RNA-seq and de novo assembly. Our results not only validate and improve the phylogenetic relationships of Nicotiana species, but also lay a foundation for analyzing key genes in leaves involved in the pathway of nicotine.
