We examined by single cell RNA sequencing the heterogeneity and progeny of NKX6.1 and/or NEUROG3 expressing cells produced in vitro by the differentiation of hiPSC cells We generated an hiPSC line where NEUROG3 is fused to a cleavable mCherry reporter and NKX6-1 to a cleavable GFP reporter (NKX6.1-GFP/NEUROG3-HA-mCherry). We differentiated the NKX6.1-GFP/NEUROG3-HA-mCherry hiPS cells towards the islet lineage until the pancreatic endocrine progenitor stage at day 13, FACS sorted the differentiated cells and sequenced the different fluorescent populations using the single-cell RNA sequencing method SortSeq.
