STARR-seq assay tiling across putative hominoid-specific enhancers and for orthologs from 11 primates and 9 ancestral reconstructions of each active tile Overall design: Three 60mm plates, each with about 1.5 million HepG2 cells, were transfected with 5ug of either our tiling library or ortholog library and 2.5ug of a puromycin expressing palsmid. Cells were selected with 1ng/mL puromycin for 24 hours and then harvested for RNA and DNA extraciton.
