Shown is a TEM image of a single ∆sor4 mucocyst docked at the plasma membrane. In contrast to the elongated mucocysts of wild-type cells, ∆sor4 mucocysts are short and round. Cells grown to stationary phase were fixed in 2% glutaraldehyde, 1% sucrose and 1% osmium at 25°C in 0.1M sodium cacodylate buffer and section-stained with uranyl acetate and lead citrate post embedding. Thin sections were viewed on a FEI (Hillsboro, OR) Tecnai G2 F30 Super Twin microscope.
