This dataset contains RNA sequencing data from sorted mouse skin antigen-presenting cells (type I conventional dendritic cells, cDC1s) and CD8? T cell subsets (tissue-resident memory T cells and effector memory T cells) collected following viral reinfection and pDC depletion (anti-PDCA1 treatment). Mice were previously vaccinated with vaccinia virus (VACV) and challenged with viral peptides in the presence or absence of anti-PDCA1 antibody to deplete pDCs. RNA-seq libraries were generated using the NEBNext Single Cell/Low Input RNA Library Prep Kit for Illumina and sequenced on a NextSeq 2000 platform, yielding single-end 61 bp reads. The dataset supports the findings reported in the manuscript “pDCs amplify tissue-resident memory CD8? T cell responses during viral reinfection,” which has been accepted for publication in the Journal of Experimental Medicine (JEM). The data can be used to explore the transcriptional impact of pDC depletion on local antiviral immune responses.
