Abstract*: Here we propose to use Capture Hi-C to generate a high-resolution structure of a whole human chromosome. As a starting point to establish Chromosome Capture Hi-C, we propose to capture the ensemble of chromosomal contacts made by human chromosome 21 in IMR90 cells (primary human fibroblasts, Encode tier 2) at unprecedented resolution. Human chromosome 21 has previously been used as a ‘model chromosome’ in pioneer transcriptome and chromatin immunoprecipitation (ChIP) studies 5. In addition, genome-wide Hi-C data for IMR90 fibroblasts are available. This data set, although affording a significantly lower resolution compared to what we aim to obtain using CCHi-C, will allow us to ‘benchmark’ our data using an independently generated chromosomal interaction data set.
