This dataset contains the transient visible absorption (visTA) data from femtoseconds to nanoseconds of the heterodimeric protein FccAB from Thiocapsa roseopersicina upon excitation at three different actinic pumps (360, 400, and 560 nm).
Experimental "raw" data (chirp-corrected) have been collected at ELI-ERIC and are designated as FccAB_XXX_DATA, where XXX indicates the actinic pump wavelength. The DATA files are 2D arrays of differential absorbance (in mOD) as a function of the pump-probe time delays (in ps, indicated in the first column), and wavelengths (in nm, indicated in the first row). These datasets are plotted in Fig. 5 and Fig. S3A.
The datasets have been analyzed by two different approaches using custom scripts: lifetime distribution analysis (LDA) without taking into account the impulse response function (IRF), and global kinetic analysis (GKA) with a sequential model and fitted Gaussian IRF.
LDA outputs lifetime density maps (FccAB_XXX_LDA_LDM files, Fig. S3B), average dynamical contents (FccAB_XXX_LDA_ADC files, Fig. S6A), and decay-associated difference spectra (FccAB_XXX_LDA_DADS files, Fig. S6B). The LDM files obtained by inverse Laplace transform using the maximum entropy method are 2D arrays of the amplitudes as a function of the wavelength (in nm, indicated in the first column), and the lifetimes (in seconds, indicated in the first row). The ADC files report the normalized ADC as a function of the lifetime (in seconds, indicated in the first column). The DADS files are 1D arrays of the differential absorbance (in mOD) as a function of the wavelength (in nm, indicated in the first column). Columns 2 and 3 represent components 1 and 2 (lifetimes in seconds indicated on top).
GKA outputs the species fraction (FccAB_XXX_GKA_FRACTION files, Fig. S5A), and evolution-associated difference spectra (FccAB_XXX_GKA_EADS files, Fig. S5B). In FRACTION files, the first column indicates the time delays (in s), the second column indicates the IRF, and the third and fourth columns are the species fraction of components 1 and 2 (lifetimes indicated on top). In the EADS files, the first column indicates the wavelength (in nm), and the second and third columns report the differential absorbance (in mOD) of components 1 and 2 (lifetimes in seconds indicated on top).
