To examine the candidate genes for GWAS at the transcriptional level, we selected 40 individuals each of EPA+DHA extreme phenotypes in the F2 population for mixed-pool transcriptome sequencing In this study, a total of 458 oysters from the F2 population resulting from the crossbreeding of C. angulata and C. gigas were utilized for liquid microarray genotyping. The F1 population were established through the crossbreeding of the 30 female C. angulata from Zhangzhou, Fujian (117°41'35 "E, 23°56'42 "N) and 30 male C. gigas from Jiaonan, Qingdao (119°57'3 "E, 35°43'20 "N), involving 30 or more oysters for both male and female parents. The F2 population was subsequently produced by the mating of the 30 female and 30 male F1 individuals and then cultured in Jiaonan, Qingdao, China. At the age of 11 months, a total of 458 F2 individuals were sampled in situ.Two extreme phenotype groups, H (high EPA+DHA) and L (low EPA+DHA), were selected from the GWAS population for transcriptomic analysis. Each group consisted of a total of 40 oyster individuals, and their total RNA was mixed equally for three technical replicates.
