Gene expression profiling of sigmoid colonic tissue form patients with ulcerative colitis and healthy controls

To assess mRNA gene expression levels in patients with ulcerative colitis (UC) and healthy controls. Two sigmoid colonic biopsies in close proximity were collected from patients with ulcerative colitis (n=59) and controls (n=19) during endoscopy. RNA from each biopsy was purified and analysed seprately using the Affymetrix HGU219 arrays on 96-PEG plates where 100 ng of RNA was labelled using the 3-IVT expression kit and hybridized to the HGU219 chip. Disease activity was assed by the Mayoscore (Schroeder et.al Coated Oral 5-Aminosalicylic Acid Therapy for Mildly to Moderately Active Ulcerative Colitis.
