Exploring the potential mechanism of Zhuanggu Zhitong Capsule in the treatment of osteoporosis in ovariectomized rats based on the transcriptome

Objective: To analyze the differentially expressed genes and related pathways of Zhuanggu Zhitong Capsule (ZGZTC) in the treatment of osteoporosis in ovariectomized rats based on transcriptome sequencing. Methods: Eighteen SD female rats were randomly divided into sham operation group, model group and ZGZTC group, each with 6 rats. Intervention was started 1 week after the model was established. The experimental group was given ZGZTC (567mg/kg), and the sham operation group and model group were given (1ml/150g) volume of distilled water, once a day for 13 consecutive weeks. On the second day after the last administration, samples were taken and blood was collected from the abdominal aorta to prepare the corresponding group of serum. RNA-Seq sequencing was performed to screen differentially expressed genes and KEGG function Annotation analysis and GO enrichment analysis.
研究目的:本研究旨在基于转录组测序(Transcriptome Sequencing)技术,分析壮骨止痛胶囊(Zhuanggu Zhitong Capsule,ZGZTC)治疗去卵巢骨质疏松模型大鼠的差异表达基因(Differentially Expressed Genes)及相关调控通路。实验方法:将18只雌性Sprague-Dawley(SD)大鼠随机分为假手术组、模型组与ZGZTC给药组,每组6只。造模1周后开始干预:给药组给予壮骨止痛胶囊(567mg/kg)灌胃,假手术组与模型组给予每150g体重对应1ml体积的蒸馏水灌胃,每日1次,连续给药13周。末次给药次日,采集腹主动脉血液以制备各组血清样本,随后进行RNA-Seq测序以筛选差异表达基因,并开展京都基因与基因组百科全书(KEGG)功能注释分析及基因本体(Gene Ontology,GO)富集分析。
