Flow cytometry data supporting "Lectin Flow Cytometry for Cell Surface Glycan Profiling: An Accessible Approach to Glycophenotyping"

This dataset contains the original flow cytometry data (FCS files) supporting the experimental results presented in the Technical Note entitled “Lectin Flow Cytometry for Cell Surface Glycan Profiling: An Accessible Approach to Glycophenotyping.” The data were generated using murine splenocytes and illustrate key methodological considerations for implementing lectin-based glycophenotyping in multiparametric flow cytometry. The experiments include lectin titration, assessment of lectin binding following cellular activation, comparison of fluorochrome and detection strategies, competitive inhibition assays, and integration of lectin staining with viability assessment, conventional immunophenotyping, and CFSE-based proliferation analysis. The dataset corresponds to Figures 1A–D and 2A, 2B, and 2D of the manuscript. Figure 2C reproduces previously published data from Sierra-Ulloa et al. (2024) and is therefore not included in this repository. These files are provided to support transparency, reproducibility, and independent examination of the flow cytometry experiments described in the manuscript.
