To understand the molecular changes driving the myeloid-biased hematopoiesis and the potential of leukemia evolution by restricting the differentiation and apoptosis in HSPCs of OP-HU mice, single cell RNA sequencing was performed to analyze the transcriptional changes of Lin-/c-kit+ HSPCs from the femur and tibia of OP-HU and control mice. Overall design: Lin-/c-kit+ HSPCs from the femur and tibia of OP-HU and control mice were enriched repectively by combination of lineage depletion kit (Miltenyi Biotech, 130-090-858) and CD117 MicroBeads (Miltenyi Biotech, 130-091-224). Lin- CD117+ enriched HSPCs were dispelled into single cell for further single-cell RNA sequencing analysis (Singleron Biotech Co., Ltd, Nanjing, China)
