This study was performed with the goal of understanding the roles of Leishmania donovani SET7 and SET29 proteins. Using homologous recombination to knock out set7 and set29 genes, set7-null (set7-/-) and set29 (set 29-/+ and set29-/-/+) mutant parasites were created. This was followed by whole genome sequence analyses of wild type, set7-/-, set 29-/+ and set29-/-/+ parasites. Our data reveals, that in wild type parasites almost all the chromosomes are disomic except chromosome 26 which is trisomic and chromosome 31 which is tetrasomic. The set7-null mutant exhibited somy changes of 5 chromosomes (2, 8, 11, 26, and 29). The two set 29 mutants exhibited somy changes of 2 chromosomes.
